Wortmannin as a unique probe for an intracellular signalling protein, phosphoinositide 3-kinase

Abstract

Wortmannin is a fungal metabolite that so far has been shown to act as a selective inhibitor of phosphoinositide 3-kinase. It can therefore be used to investigate the convergence between two major cellular signalling systems: those involving G-protein-coupled receptors and those involving receptor tyrosine kinases. Importantly, wortmannin can enter intact cells, making whole-cell studies of the above signalling pathways possible.

      For allowing the inhibitors to penetrate the cells and avoid eventual interaction between the compounds, the inhibitors were added 20 min before the inducers. The cells were treated with the PLD inhibitor1-butanol in concentration of0.01, 0.05 and 0.1%, and separately with 1, 10, and 100 μM WTM which in addition to its PI3K inhibiting activity is known also as autophagy inhibitor (Ui et al., 1995; Blommaart et al., 1997; Walker et al., 2000; Ueno et al., 2016). The involvement of ROS was studied by applying compounds possessing ROS scavenging properties: L-galactonic acid-γ-galactone (L-gal, immediate precursor of ascorbic acid), kaempferol (KP, a plant flavonoid) and -N-acetyl-L-cysteine (L-NAC, a by-product of glutathione).
