Similar to PDGF in both structure and activation paradigm, the VEGF family also belongs to the cysteine knot superfamily of signalling molecules (Tischer et al., 1989), where VEGF-A, the most abundant and active member, activates VEGFR2 found on endothelial cells to initiate angiogenesis and vasculogenesis (Apte et al., 2019; Simons et al., 2016). VEGF-A can signal directly through both PDGFRα and PDGFRβ to increase mesenchymal stem cell proliferation and migration (Ball et al., 2007).

The amino acid sequences predicted by cDNA cloning showed that the homology of VEGF with the A and B chains of human platelet derived growth factor was about 21% and 24%, respectively [163]. During embryonic vascular development, VEGF-A is the most extensively studied member of the VEGF family and expressed by the extraembryonic visceral endoderm as early as embryonic day (E) 7.5, which is consistent with blood island formation in the yolk sac [164].
