Indeed, MCF-7 breast tumor cells exhibited a disruption of mitochondrial respiration after its treatment with melatonin [165]. Curiously, it has been demonstrated in pituitary prolactin-secreting tumor that melatonin disrupts the activities of the four mitochondrial complexes, leading to a mitochondrial dysfunction and the induction of apoptosis [166].

Oxygen consumption was measured as described previously (16) in mclk1 +/+, +/−, and −/− ES cells. To further study the function of CLK-1 and the biological roles of ubiquinone, we disrupted the mclk1 locus in murine ES cells by homologous recombination and produced heterozygous and homozygous mice using standard methods (see “Experimental Procedures” and Figs. 1 and 2, c andd).
